Aberrant TDP‐43 phosphorylation: a key wind gap from TDP‐43 to TDP‐43 proteinopathy

نویسندگان

چکیده

TDP‐43 proteinopathy is a kind of neurodegenerative diseases related to the TAR DNA‐binding protein 43‐kDa molecular weight (TDP‐43). The typical include amyotrophic lateral sclerosis (ALS), frontotemporal lobar degeneration (FTLD), Alzheimer's disease (AD), Parkinson's (PD) and so on. As process cannot be blocked or slowed down, these patients have poor quality life prognosis, bring huge burden family society. So far, specific pathogenesis not clear, there no effective preventive measure treatment program for this disease. plays an important role in triggering promoting occurrence progression proteinopathy. hyperphosphorylation undoubtedly factor Hyperphosphorylation can inhibit degradation TDP‐43, aggravate aggregation protein, increase wrong localization cells, enhance cytotoxicity TDP‐43. More more evidences show that Inhibition may one strategies Therefore, article reviews phosphorylation mechanisms.

Expression of TDP43 in cervical cancer

TDP43 is ubiquitously expressed in human tissues and plays a crucial role in a wide range of neurodegenerative diseases. However, the study in cancer especially in cervical cancer is less known. The aim of this study is to identify the expression and function of TDP43 in cervical cancer. Here we reported that protein and mRNA level of TDP43 expression was up-regulated in cervical cancer tissues...

[Molecular pathogenesis of ALS in TDP43 era].

To clarify the molecular pathogenesis of amyotrophic lateral sclerosis (ALS) associated with TAR-DNA binding protein 43 kDd (TDP-43), the quality and quantity of TDP-43 take a crucial role. Regarding to the quality of TDP-43, TDP-43 has been reported as an aggregate-prone protein. Especially the C-terminus of the TDP-43 tends to form aggregate and has prion-like domain. Interestingly the mutati...

Widespread RNA metabolism impairment in sporadic inclusion body myositis TDP43-proteinopathy☆

TDP43 protein mislocalization is a hallmark of the neurodegenerative diseases amyotrophic lateral sclerosis and frontotemporal dementia, and mutations in the gene encoding TDP43 cause both disorders, further highlighting its role in disease pathogenesis. TDP43 is a heterogenous ribonucleoprotein, therefore suggesting that alterations in RNA metabolism play a role in these disorders, although di...

Aberrant Localization of FUS and TDP43 Is Associated with Misfolding of SOD1 in Amyotrophic Lateral Sclerosis

BACKGROUND Amyotrophic lateral sclerosis (ALS) is incurable and characterized by progressive paralysis of the muscles of the limbs, speech and swallowing, and respiration due to the progressive degeneration of voluntary motor neurons. Clinically indistinguishable ALS can be caused by genetic mutations of Cu/Zn superoxide dismutase (SOD1), TAR-DNA binding protein 43 (TDP43), or fused in sarcoma/...

Structural Dynamics of Human Argonaute2 and Its Interaction with siRNAs Designed to Target Mutant tdp43

The human Argonaute2 protein (Ago2) is a key player in RNA interference pathway and small RNA recognition by Ago2 is the crucial step in siRNA mediated gene silencing mechanism. The present study highlights the structural and functional dynamics of human Ago2 and the interaction mechanism of Ago2 with a set of seven siRNAs for the first time. The human Ago2 protein adopts two conformations such...
